I was downloading a file, changed the BT lock setting to never lock, then let it finish.

The file finished fine but it's permanently locked. It can't be moved, deleted, or renamed. It ALWAYS says the file is in use, even when I'm not on the LAN and in safemode.

I can copy the file, but it results in the same thing, the newly created file cannot be moved, renamed, or deleted.

How can I manually unlock the file??

Let me guess, it's a video file? Windows Explorer can lock up on video files it has trouble with, locking them. Check your codecs.

If updating your codecs doesn't help, you can access the file through the console (cmd/command) if you're familiar with it. But do this before using windows explorer (or kill explorer task on W2k/XP if already locked).
You can also try to change the FourCC, if that's the cause of the problem.
